Xtandi (enzalutamide) blocks androgen signaling in prostate cancer. It prevents androgens such as testosterone and dihydrotestosterone from activating the androgen receptor, which slows tumor growth driven by androgen signaling.[1]
Xtandi acts as an androgen receptor inhibitor. It binds the androgen receptor and prevents it from signaling in key steps of the pathway, including receptor activation and downstream transcription of androgen-responsive genes.[1]
By shutting down androgen receptor signaling, Xtandi reduces the growth and survival signals cancer cells rely on. This is why it is used in patients with advanced prostate cancer, including cases where the disease has progressed despite earlier androgen deprivation therapies.[1]
Unlike treatments that lower androgen production (androgen deprivation therapy), Xtandi targets the receptor signaling step directly, stopping the androgen receptor from functioning even when androgens are present.[1]
