Common Side Effects of Tigecycline
Tigecycline, an intravenous antibiotic for complicated infections, often causes nausea (26% of patients), vomiting (18%), and diarrhea (13%). Other frequent effects include headache (13%), abdominal pain (11%), and pain at the injection site (9%). These typically occur early in treatment and may resolve without stopping the drug.[1][2]
Serious Side Effects and Warnings
Black box warnings highlight increased mortality risk (4% vs. 3% in comparators), mainly from worsening infections or sepsis—avoid in non-severe infections. Pancreatitis occurs in about 1-2% of cases, with severe abdominal pain signaling it. Hypersensitivity reactions, including anaphylaxis, and superinfections like C. difficile colitis are possible. Hepatotoxicity and fetal harm in pregnancy (Category D) also require monitoring.[1][2]3
Side Effects by Body System
- Gastrointestinal: Beyond nausea/vomiting, dyspepsia (8%) and constipation (4%).
- Laboratory Changes: Elevated liver enzymes (ALT 22%, AST 20%), increased lactate dehydrogenase (15%), and amylase (7%); monitor in liver disease patients.
- Hematologic: Anemia (7%), leukocytosis (7%).
- Other: Dizziness (8%), fever (5%), rash (4%). Tooth discoloration and enamel hypoplasia noted in animal studies, with human cases post-marketing.2
Who Experiences More Side Effects?
Patients with ventilator-associated pneumonia or severe renal/hepatic impairment face higher risks. Discontinuation due to adverse events happens in 5-10% of cases, mostly GI-related. Elderly patients show similar profiles but higher mortality risk.[1][2]
Managing Tigecycline Side Effects
Start with antiemetics for nausea. Infuse slowly (30-60 minutes) to reduce vein irritation. Regular blood tests track liver function and pancreatitis markers. Report severe symptoms immediately; drug may need switching.3
