How to Administer Ozempic
Ozempic (semaglutide) is a once-weekly subcutaneous injection for type 2 diabetes management and weight loss. Use the prefilled Ozempic pen. Store unused pens in the refrigerator (36–46°F or 2–8°C); pens in use can stay at room temperature (59–86°F or 15–30°C) for up to 56 days.[1]
Preparing the Dose
- Check the pen label for your prescribed strength (0.25 mg, 0.5 mg, 1 mg, or 2 mg).
- Wash hands. Inspect the solution—it should be clear and colorless. Do not use if cloudy or discolored.
- Attach a new needle (included or compatible NovoTwist). Remove paper tab and needle caps.
- Turn the dose selector to the flow check symbol (looks like two dots). Hold pen upright and press button until a drop appears at needle tip—repeat up to 6 times if no drop.[1][2]
Injection Sites and Technique
Inject under the skin in the abdomen (at least 2 inches from navel), thigh (front), or upper arm. Rotate sites weekly to avoid irritation.
- Clean site with alcohol swab; let dry.
- Pinch skin gently. Insert needle straight in at 90-degree angle.
- Press and hold dose button fully until dose counter shows 0 and beep (if applicable). Count to 6 slowly while holding to ensure full dose.
- Release skin, remove needle straight out. Do not rub site.[1][2]
Timing and Scheduling
Inject once weekly, any time of day, with or without food. Start with 0.25 mg for 4 weeks (dose escalation), then increase as prescribed. If missed, take within 5 days; otherwise skip and resume next scheduled dose.[1]
Common Mistakes to Avoid
Do not reuse needles—discard in sharps container. Never inject into muscle, vein, or irritated skin. Prime pen every time before dosing. If pen is dropped or damaged, use a new one.[2]
Dose Escalation for Therapy
| Week | Dose | Purpose |
|------|------|---------|
| 1–4 | 0.25 mg | Initiation, minimize GI side effects |
| 5+ | 0.5 mg (or higher per doctor) | Maintenance; max 2 mg weekly |
Consult your doctor for personalized adjustments, especially if side effects like nausea occur.[1]
