Does vyvanse affect serotonin?

Short answer: Vyvanse (lisdexamfetamine) mostly affects dopamine and norepinephrine. It can cause some serotonin release, but that's not its primary action and the effect on serotonin is relatively small at typical doses.

- Lisdexamfetamine is a prodrug of dextroamphetamine. It increases monoamines by promoting release from neurons and, to a lesser extent, blocking reuptake.
- The strongest effects are on dopamine (DAT) and norepinephrine (NET). Serotonin (SERT) is affected to a lesser extent.

- Serotonin syndrome is unlikely with lisdexamfetamine alone, but it can be a concern if combined with other serotonergic medications (SSRIs, SNRIs, MAOIs, TCAs, tramadol, triptans, etc.). Be aware of symptoms such as agitation, confusion, rapid heart rate, high blood pressure, dilated pupils, loss of coordination, or muscle rigidity, and seek urgent care if they occur.
